FBI agents keep out of Benghazi

By The Associated Press

For security reasons, FBI agents are staying away from the Libyan city where a U.S. ambassador and three other Americans were killed, two law enforcement officials said Friday.

The officials say the bureau is not going to put agents in harm's way and that the city of Benghazi must be made secure before the FBI sends investigators there.

The officials demanded anonymity because they are not authorized to speak on the record about an ongoing investigation.

FBI agents were sent to Libya last week to look into the Sept. 11 attack that killed Ambassador Chris Stevens, a State Department computer expert and two former U.S. Navy Seals.

Terrorist groups in Libya tried to coalesce in month leading to consulate attack, officials say

At FBI headquarters Friday, spokesman Paul Bresson said "we are moving forward with our investigation," but Bresson declined to comment on the specific location of the agents.

Separately, the State Department is further reducing the U.S. Embassy staff in Tripoli for security reasons. The embassy warned Americans of possible demonstrations in the capital and Benghazi on Friday.

On Thursday, Libya's leader said his government had disbanded about 10 militia groups and will continue to take action against Muslim extremists.

President Mohammed el-Megarif said the attack on the U.S. Consulate earlier this month that killed the four Americans was a final straw. He did not say when the militias were disbanded, or how many remain.

Tourists headed for Everest region among 19 killed in fiery Nepal ...

Nineteen people have died in a plane crash in Nepal. They were on their way to climb Mount Everest. The plane crashed into a field shortly after take-off from the capital Kathmandu. It was bound for Lukla, the starting point for a trek through the Himalayan mountains to the base camp of Mount Everest. ITV's Paul Davies reports.

By NBC News wire services

KATMANDU, Nepal -- A plane carrying trekkers to the Everest region crashed and caught fire just after takeoff Friday in Nepal's capital, killing 19 people.

The victims included British, Chinese and Nepali passengers, authorities said.

The pilot of the domestic Sita Air flight reported trouble two minutes after takeoff, and Katmandu airport official Ratish Chandra Suman said the pilot appeared to have been trying to turn back.?

The crash site is only 547 yards from the airport, and the wrecked plane was pointing toward the airport area.

Reuters said it was a twin-engine, propeller-driven Dornier aircraft.

Investigators were trying to determine the cause of the crash and identify the bodies. Suman said he could not confirm if the plane was already on fire before it crashed.

Villagers forced back by flames
Cellphone video shot by locals showed the front section of the plane was on fire when it first hit the ground and it appeared the pilot had attempted to land the plane on open ground beside a river.

The fire quickly spread to the rear, but the tail was still in one piece at the scene near the Manohara River on the southwest edge of Katmandu.

Villagers were unable to approach the plane because of the fire and it took some time for firefighters to reach the area and bring the fire under control.

A plane carrying 19 people crashed shortly after taking off in Katmandu, Nepal, catching fire and killing all on board. NBCNews.com's Dara Brown reports.

Nepal officials: 6 survive, 15 killed as plane hits mountain in Himalayas

Soldiers and police shifted through the crash wreckage looking for bodies and documents to help identify the victims.

Seven passengers were British and five were Chinese; the other four passengers and the three crew members were from Nepal, authorities said.

Large numbers of local people and security forces gathered at the crash site. The charred bodies were taken by vans to the hospital morgue.

Gateway to Everest
The weather in Katmandu and surrounding areas was clear on Friday morning, and it was one of the first flights to take off from Katmandu's Tribhuwan International Airport. Other flights reported no problems, and the airport operated normally.

The plane was heading for Lukla, the gateway to Mount Everest. Thousands of Westerners make treks in the region around the world's highest peak each year. Autumn is considered the best time to trek the foothills of the Himalayan peaks.

93% Farewell, My Queen

If you are expecting a French film chronicling the last days of Marie Antoinette, then you may be in for a slight disappointment. Brilliantly directed by the Benoit Jacquot (who also co-wrote the much less impressive script) and starring Diane Kruger as Marie Antoinette and Lea Seydoux, who plays one of the Queen's readers named Sidonie Laborde, and is also the protagonist of this film (for some reason) "Farewell, My Queen" is advertised as the story of the last days of Marie Antoinette, but it's more like the somewhat muted story of the two months before the "last days" of Marie Antoinette. So, even though "Farewell, My Queen" is a better movie than Sofia Coppola's ill advised, indie rock inspired, "Marie Antoinette", due to the faux-final-days story structure, in conjunction with the fact that "Farewell, My Queen" uses Marie Antoinette as a side character, telling the story from the point of view of Sidonie Laborde (a nobody) this is a film which may have many audiences scratching their heads as to why the need to show this particular moment in the life of Marie Antoinette. As I alluded to above, "Farewell My Queen" chronicles a miniscule portion in time during the last months of Marie Antoinette's reign. However, audiences are not treated to "the good part", or the part they undoubtedly came to see. What I mean by this is there are almost no visuals of the actual Revolution, plus (and more importantly) we don't actually get to witness the final days of Marie Antoinette's life simply because "Farewell, My Queen" curiously plays out through the eyes of one of her ladies-in-waiting. Instead, audiences are forced to sit through a pseudo-love story that focuses not on the very interesting end of Marie Antoinette's reign, but on the minutiae right before it! This film is the equivalent of sitting through the love story of Jack and Rose, with the movie ending just before the Titanic strikes the iceberg. Yes, I am aware that this is a script which was adapted from a critically acclaimed novel by Chantal Thomas, BUT it still doesn't make the story's focus any less misguided. I mean, there are some scenes which do attempt to create a somewhat intriguing love story, an aspect which must be fully accredited to some fantastic mood setting by Jacquot, but in the same vein, this film never rises above said simplistic love story. Side Note: Another issue many audiences will likely come across is how frivolously "Farewell, My Queen" throws its viewers right into the deep end of this story, with little exposition. In fact, the greatest individual flaw which hinders "Farewell, My Queen" will be seen in the audiences immediate realization that neither Jacquot's visuals or the engaging performances (which I will speak about later) are going to give those not formally versed in the players of the French Revolution and the fall of Versailles, the background information they may desperately desire, as this film half-introduces more and more characters of seeming importance. So, do yourself a favor, if you are going to see "Farewell, My Queen" (at the very least) peruse the Marie Antoinette Wiki page before going to see this film. With that said, the visuals (the set design in conjunction with the director) are somewhat breathtaking at times, due to Jacquot making some very brave directorial choices, including tons of long takes which trail behind characters as they weave in and out of crowds, giving audiences an intimate feeling of the atmosphere of late 1700's France, and a few beautifully constructed shots of the landscapes. And I guess it doesn't hurt that the two female leads, Seydoux and Kruger, both give engaging performances. But even if Kruger's interpretation of Antoinette is one of the best I've ever seen and Seydoux is so captivating to watch as she effortlessly takes control of the movie every time she is on screen, there is simply not enough in the story or the writing to give reasoning to the eccentric behavior or motives behind the actions of Marie Antoinette or (more importantly) give a reason as to why Sidonie is so infatuated with her. So, even though, in the latter half of the film (more than an hour in) Jacquot does create an atmosphere which allows these characters to somewhat blossom, many will find it hard to care about a Marie Antoinette story that contains no beheadings. Final Thought: Based on a "last days" plot which attempts to imitate a much better film like "Downfall", even with some spectacular direction and two engaging female performances, "Farewell, My Queen" is nothing more than this year's "My Week With Marilyn", telling a story which focuses on characters nobody really cares about, rather than simply creating a storyline around the life/last days of (in this case) Marie Antoinette. So, even if you are a Marie Antoinette fan, with the overall structure the way it is, "Farewell, My Queen" is nothing more than DVD worthy at best. ?To Whom Are You Referring?? vs. ?Who are you talking about ...

?To whom are you referring?? vs. ?Who are you talking about?? Writing the Way People Talk

Sep 27, 2012

?

I make the bulk of my living as a corporate copywriter and freelancer. That means I spend a lot of time writing spam and junk mail.

Oh, I could tell you I write ?marketing collateral? or I ?develop B 2B and B2C messaging campaigns? but unless you?re actually in marketing and advertising, that?s probably not going to mean much to you. But when I say, ?spam? and ?junk mail? general folks I bump into on the street instantly know what I?m talking about. We have a baseline. I can refine from there.

I was thinking about the ways people talk recently when I received some feedback about one of my projects. Someone pointed out that the grammar of the subject line in an email I wrote was incorrect.

The original was something along the lines of: ?Found who you?re looking for??

As most copyeditor folks and even some high falutin? writers will tell you, that sentence commits a number of sins. If that sentence were written in a grammatically correct way, it would read: ?Have you found the person for whom you are looking??

Catchy, isn?t it? Obviously, I should have used that one.

Right?

?Have you found the person for whom you are looking?? may be grammatically correct, but it fails to do what I need it to do. As an email subject line, it?s too long. It also?sounds awkward and off-putting, distancing. It uses a ?high? narrative language that implies I?m speaking down to somebody, and that?s not my intent -?I want to speak in a simple,?relatable way.

Yes, I thought about this.

Writing the way people talk, in a way they?ll find accessible, is as much a writing skill as ?knowing the rules.? I make the same sorts of tough choices in my fiction writing, too. My characters use references pertinent to their world and worldview, education and training. Having a good ear for the way people talk ? without being a literal transcriptionist (cause whoa boy nobody would understand a thing) is a great skill for a writer.

Most people don?t speak in a grammatically correct way. In fact, when we hear grammatically correct stuff like, ?Discover the?teacher for whom you are looking,? it sounds a little funny. That?s because it often doesn?t gel with our internal grammar. Our internal grammarian is always going to be a jump ahead of the printed ?rules.?

How many words do you use every day that aren?t even in the dictionary? How many times have you used the word ?whom? this week? I spend a lot of time thinking about?the audience?I?m writing for, and how those folks are used to communicating with people. Again, this pertains to fiction, too. There?s a reason that there are more people reading simply-written books with?solid but simple stories?than purple-prose doorstoppers.

That?s not to say I?m against lovely language ? I prefer weird, gorgeous language, myself- but my fiction audiences and my freelancing/corporate audiences are very different, and I write differently for each.

When writers tell you to ?learn all the rules? and then break them,? grammar is one of those rules. You should know what you?re supposed to say, sure ? because then you can make a conscious choice to actually say what you mean in a way your audience will understand.

WHY IT MATTERS: Gun control

The issue:

Frightening episodes of gun violence have been splayed across front pages with alarming frequency this campaign season: the movie theater killings in Colorado, the Sikh temple shootings in Wisconsin, the gun battle outside the Empire State Building, and more. Guns are used in two-thirds of homicides, according to the FBI. But the murder rate is less than half what it was two decades ago.

___

Where they stand:

Neither President Barack Obama nor Republican Mitt Romney has had much to say about guns during the campaign, except when prodded by high-profile events such as the Colorado shootings. Obama hasn't pushed gun control measures as president, and Romney says he doesn't think the nation needs new gun laws. Both style themselves as defenders of the Second Amendment right to bear arms.

Obama has signed laws letting people carry concealed weapons in national parks and in checked bags on Amtrak trains. He's voiced support for a renewed ban on assault-type weapons but hasn't tried to get that done. He blames Congress for opposing such measures.

Romney suggested after the Colorado shootings that he favors tougher enforcement of existing gun laws. He said he doubts whether new laws would help. The key, he said, is to identify deranged or distressed people and then "keep them from carrying out terrible acts." It's a big shift in tone from his days as Massachusetts governor, when he vowed to protect the state's "tough gun laws" and signed a ban on assault weapons.

___

Why it matters:

Don't expect gun control to be high on the presidential agenda no matter who wins in November. Although Obama is more open to gun ownership restrictions than Romney, neither seems inclined to push it. That's not to say it makes no difference who gets elected when it comes to gun control. Supreme Court appointments could make a big difference.

But it has faded as a political issue. Crime just isn't the public concern it was years ago. And it's getting harder to make the argument that stricter gun laws are needed when violent crime has been decreasing without them.

Although high-profile shootings inevitably stir up talk about tighter gun controls, over time the public has shifted in favor of the right to own guns and away from increased limitations on gun ownership.

In 1990, 78 percent of Americans said laws covering the sale of firearms should be stricter, and 19 percent said they should remain the same or be loosened, according to a Gallup poll. By the fall of 2004, support for tougher laws had dropped to 54 percent. In last year's sounding, 43 percent said they should be stricter, and 55 percent said they should stay the same or be made more lenient.

Still, odds are good that the next president will have a chance to fill at least one Supreme Court seat, and the current court is narrowly divided on gun-control questions. In a 5-4 vote two years ago, for example, the court held that Americans have the right to own a gun for self-defense anywhere they live, expanding the conservative court's embrace of gun rights. The court left unresolved questions about what kinds of gun-control laws fit comfortably with the Second Amendment.

An Obama appointee could be expected to be friendlier to gun controls than would a Romney nominee.

Obama could decide to push for an assault weapons ban in a second term, when he wouldn't have to worry about re-election repercussions. But it still would be unlikely to advance through a House that is expected to remain firmly in Republican hands.

Will unbeaten Barca win La Liga?

It wasn?t easy, but thanks to a late Xavi Hern?ndez goal, Barcelona beat Granada this weekend, running their?La Liga record to a perfect 15 points in five games. What does that kind of start mean for Bar?a?s chances to reclaim Spain?s title?

As AS tells us (taking the data from Twitter?s Mister Chip), this is the seventh time Barcelona have been perfect through five rounds. On four of those occasions, Bar?a went on to win the title.

Is that a good ratio? Or what you?d expect from teams who start perfect through five rounds? Somebody needs to write a script that runs those scenarios. We don?t know. The stat sounds interesting though not very informative about 5-0-0 starts in general.

So AS/Mister Chip looked at more 5-0-0 starts. A team has started 5-0-0 on 24 other occasions in?La Liga history. Ten of those sides went on to win the title, a much lower success ratio than Barcelona?s. No team has started 5-0-0 and finished outside the top four.

It?s not much, but the 31 cases gives us some perspective. Perhaps it doesn?t tell us anything useful, but it hints there might not be anything useful to be told.

But even without looking at past data, we dig a little (only a little) deeper on Barcelona?s season to see if we can find something more descriptive than just ?perfect start?:

  • Barcelona seems to have played an pretty easy schedule, so far (Real Sociedad, Osasuna, Valencia, Getafe, Granada). None of their opponents sit higher than 14th. Another way to put it: Barcelona?s five matches have all been against teams Spain?s bottom seven.
  • However, those five teams are 4-4-6 (W-L-D) when they?re not facing Barcelona, suggesting Bar?a?s great start is making their schedule seem superficially weaker than it actually is.
  • In attack, Barcelona?s 14 goals look nice (it?s the highest total in the league), but let?s look at how Bar?a?s scoring rate compares to other sides performance against the same competition. Crunch the numbers and Barcelona?s scored 113 percent more goals (per game) than the rest of the league against those five opponents.
  • Use the same logic and look at goals allowed and Barcelona?s defense has performed 40 percent better than the rest of Spain (against those five).

The defense has been the big concern for Barcelona. Though the numbers are good (only allowing three goals in five games), Bar?a?s back line passing isn?t the eye test. This weekend against Grenada, they almost gave up a late winner after two?impetuous?Granada players lept out the windows of their parked bus. Stats help paint a more rounded picture, but when Javier Mascherano and Alex Song are allowing Fabian Orellana an uncontested shot on Victor Valdes with the game in the balance, that?s cause for concern.

And although Barcelona?s done a good job of containing their opponents, those five teams aren?t putting up numbers that suggest they?re the most meaningful tests to Bar?a?s possession-hogging style. Barcelona?s only allowing 0.60 goals per game, but against the rest of the league, their opponents have only managed one goal per game.

Beyond weddings and roses: Moon's US influence

The mass weddings at Madison Square Garden and the roses sold by eager evangelists on street corners may be the most enduring images of Rev. Sun Myung Moon's Unification Church in the United States.

But Moon's U.S. empire extended far beyond the movement and the Moonies, as Moon's followers were both affectionately and irreverently known. Moon had holdings in media, real estate, universities and even sushi sales.

A look at Moon and the Unification Church's influence around the U.S.:

___

WEDDINGS

Moon introduced the mass weddings he started in Seoul to the U.S. with a 1982 wedding at New York's Madison Square Garden, drawing over 2,000 people, all dressed alike in suits and satin dresses. In 2009, as he turned 90, the church said Moon married 45,000 people in simultaneous ceremonies worldwide in his first large-scale mass wedding in years. Tens of thousands in the U.S. participated in Moon's "blessing ceremonies," many at the Manhattan Center, an event space owned by the church.

___

MEDIA

Moon founded the Washington Times in 1982 as a conservative voice in the nation's capital. At one time, he owned newspapers in New York, Japan and South America. A church subsidiary purchased the United Press International news service over a decade ago.

Moon underwrote the operations of the Washington Times for years as an alternative to the larger Washington Post. The newspaper cultivated a generation of conservative journalists, but also struggled financially recently as it faced declining circulation along with many other newspapers nationwide.

The Times cut about 40 percent of its staff at one point and eliminated its sports section. As Moon aged and transferred control of his media empire to his children, management shake-ups followed at the Times, which also found itself in court over the religious beliefs of the Moon family. The newspaper's former opinion editor sued in 2009 claiming he was pressured by executives to attend a Unification Church event.

___

REAL ESTATE

Moon's holdings include The New Yorker Hotel, an Art Deco landmark near Penn Station in midtown Manhattan; the Manhattan Center, with event spaces that include The Hammerstein Ballroom. The church owned several estates and buildings in Irvington and Tarrytown, New York, north of the city, including Moon's 17-acre (seven-hectare) estate.

____

UNIVERSITIES

The Unification Church rescued the financially troubled University of Bridgeport in Connecticut, which nearly closed in 1991 because of money problems. The Professors World Peace Academy, which was funded by the church, gave $110 million to the school over a dozen years ending in 2003.

At the time, Moon and the academy envisioned creating a network of universities around the world, with each campus open to students and faculty of the others. In 1995, Moon visited UB for the first time and gave a nearly hour-long speech as he received an honorary degree. Opponents, dubbing the school "Moonie U.," protested outside.

University spokeswoman Leslie Geary said Sunday that school officials were thankful for the church's support.

"The Rev. Moon supported the university during a difficult time," Geary said.

____

SUSHI

Moon has purchased and controlled a number of seafood companies around the world, including True World Foods, a wholesaler that distributes sushi and other seafood to more than 8,000 Japanese restaurants around the U.S. Moon has also claimed holdings of seafood and shipbuilding companies in Alabama and Alaska.

___

TAXES

Moon lived mostly in New York State, but for 13 months was an inmate doing kitchen duty at a federal prison in Danbury, Connecticut. Moon was convicted in the early 1980s of filing false income tax returns for three years in the '80s, failing to report more than $150,000 in income he deposited in New York accounts. Moon contended that the funds were being held for members of his church. He lost an appeal to the 2nd U.S. Circuit Court of Appeals, in which he was represented by constitutional law scholar Laurence Tribe.

(AP) ? When the Democratic Party's national convention opens this week, President Barack Obama's target audience won't be in the crowd. It will be the small sliver of undecided voters in battleground states who will be critical to the outcome of what the polls show is a tight race with two months to go.

Obama's campaign will turn to technology to get some of those voters engaged in the convention. The campaign will stream the entire event online, incorporate voter comments into the feed, and have convention attendees interact electronically with those watching on the Internet and mobile devices.

It's an attempt to recapture the Obama team's insurgent, grassroots appeal from 2008, when the campaign set new standards for using technology in politics. The high-tech engagement also helps the campaign capture data on voters that can be used for registration drives and get out the vote efforts.

Targeting Hispanics, the convention will be streamed online simultaneously in Spanish. Obama is hoping his sizeable advantage over Republican rival Mitt Romney with Hispanics will help him win key battleground states in the West, as well as Florida and Virginia.

Republicans also streamed much of their convention on YouTube, though the feed was not interactive.

The move to online streaming comes as television networks cut back on their convention coverage. TV viewership was down during last week's Republican convention; about 30 million people watched Romney's acceptance speech, compared to 40 million who saw Sen. John McCain accept the GOP nomination in 2008.

Starting Tuesday, a parade of high-profile speakers will stand on a blue-carpeted stage in Charlotte's Time Warner Cable Arena to vouch for Obama's economic agenda, which his team says is focused on the middle class: ending tax cuts for the rich and reducing the debt, while spending more on education, energy and infrastructure.

Several voters ? called "American Heroes" by Obama's team ? also will speak at and appear in videos at the convention, putting a human face on Obama's program.

The Democratic National Convention starts less than a week after Republicans gathered in Tampa, Fla., to nominate Romney as the party's presidential candidate. Democrats hope that by holding their convention second, Obama can emerge with momentum on his side as the race for the White House bounds into its final stretch.

Obama will largely be responsible for generating that momentum. He will close the convention Thursday night with a speech in an outdoor football stadium, just as he did in 2008. Mindful of the comparisons to four years ago, Obama's campaign is scrambling to ensure that the 74,000-seat stadium is filled to capacity. The largest crowd Obama has drawn during the 2012 campaign is about 14,000 people, far less than the jaw-dropping crowds he attracted in the 2008 campaign.

Aides say Obama won't ignore the economic woes that have defined his four years in the White House. But they say he plans to focus largely on the future, and why he believes his policies will succeed in a second term.

Obama isn't expected to outline any new policy proposals. Instead, he plans to make the case for continuing what he has started. And he is expected to double down on agenda items, such as immigration and tax reforms, that gained little traction during his four years in office.

"When the convention is over, folks with be left with a clear road map of where he thinks America needs to go," said Stephanie Cutter, Obama's deputy campaign manager. "And it will be clear what his focus will be in an Obama second term."

Working against Obama: the nation's 8.3 percent unemployment rate, sluggish economic growth and fears the economy could slip back into a recession.

There's also a general malaise. An Associated Press-GfK poll conducted earlier this month showed 60 percent of registered voters say the country is heading in the wrong direction, while just 35 percent say it is heading in the right direction.

The convention opens Tuesday with first lady Michelle Obama, whose popularity far surpasses her husband's, as a featured speaker. San Antonio, Texas, Mayor Julian Castro also is slated for that night. He will be the first Hispanic to deliver the Democratic convention's keynote address. Their roles on the convention's opening night are part of Democrats' efforts to shore up support among women and Hispanics, two crucial voting blocs where Obama holds an advantage over Romney.

Mrs. Obama is expected to make the case that Obama is the best candidate to advocate on behalf of the middle class because he has experienced their struggles himself.

Many voters already have heard Mrs. Obama's stories about her husband being raised by a single mother and his grandparents or having struggled to pay off student loans.

But she is emphasizing them again in this campaign in hopes of drawing a contrast with Romney's privileged upbringing.

Polls show voters think Obama understands the economic issues that are important to them better than Romney, even though the Republican has an edge on who voters believe is better suited to manage the economy.

Former President Bill Clinton, who is emerging as one of the campaign's most effective surrogates, will headline the convention Wednesday and formally nominate Obama. He hopes to remind voters of the flush economy he presided over and make the case that Obama's policies will lead to similar results.

Vice President Joe Biden and Massachusetts Sen. John Kerry will address the large stadium crowd Thursday night before Obama speaks.

Kerry, seen as a potential second-term secretary of state under Obama, will try to capitalize on the Democratic Party's rare advantage on national security issues. He is expected to trumpet Obama's decision to order the raid that killed Osama bin Laden and the president's plan to end the Afghanistan war, a sharp contrast to Republicans who rarely mentioned the war during their convention or the tens of thousands of troops still engaged in combat.

Obama's young daughters start school in Washington next week and are not expected to have a formal role at the gathering. But they could come to Charlotte Thursday night for their father's acceptance speech.

Design help for drug cocktails for HIV patients: Mathematical model helps design efficient multi-drug therapies

ScienceDaily (Sep. 2, 2012) ? For years, doctors treating those with HIV have recognized a relationship between how faithfully patients take the drugs they prescribe, and how likely the virus is to develop drug resistance. More recently, research has shown that the relationship between adherence to a drug regimen and resistance is different for each of the drugs that make up the "cocktail" used to control the disease.

New research conducted by Harvard scientists could help explain why those differences exist, and may help doctors quickly and cheaply design new combinations of drugs that are less likely to result in resistance.

As described in a September 2 paper in Nature Medicine, a team of researchers led by Martin Nowak, Professor of Mathematics and of Biology and Director of the Program for Evolutionary Dynamics, have developed a technique medical researchers can use to model the effects of various treatments, and predict whether they will cause the virus to develop resistance.

"What we demonstrate in this paper is a prototype for predicting, through modeling, whether a patient at a given adherence level is likely to develop resistance to treatment," Alison Hill, a PhD student in Biophysics and co-first author of the paper, said. "Compared to the time and expense of a clinical trial, this method offers a relatively easy way to make these predictions. And, as we show in the paper, our results match with what doctors are seeing in clinical settings."

The hope, said Nowak, is that the new technique will take some of the guesswork out of what is now largely a trial-and-error process.

"This is a mathematical tool that will help design clinical trials," he said. "Right now, researchers are using trial and error to develop these combination therapies. Our approach uses the mathematical understanding of evolution to make the process more akin to engineering."

Creating a model that can make such predictions accurately, however, requires huge amounts of data.

To get that data, Hill and Daniel Scholes Rosenbloom, a PhD student in Organismic and Evolutionary Biology and the paper's other first author, turned to Johns Hopkins University Medical School, where Professor of Medicine and of Molecular Biology and Genetics Robert F. Siliciano was working with PhD student Alireza Rabi (also co-first author) to study how the HIV virus reacted to varying drug dosages.

Such data proved critical to the model that Hill, Rabi and Rosenbloom eventually designed, because the level of the drug in patients -- even those that adhere to their treatment perfectly -- naturally varies. When drug levels are low -- as they are between doses, or if a dose is missed -- the virus is better able to replicate and grow. Higher drug levels, by contrast, may keep the virus in check, but they also increase the risk of mutant strains of the virus emerging, leading to drug resistance.

Armed with the data from Johns Hopkins, Hill, Rabi and Rosenbloom created a computer model that could predict whether and how much the virus, or a drug-resistant strain, was growing based on how strictly patients stuck to their drug regimen.

"Our model is essentially a simulation of what goes on during treatment," Rosenbloom said. "We created a number of simulated patients, each of whom had different characteristics, and then we said, 'Let's imagine these patients have 60 percent adherence to their treatment -- they take 60 percent of the pills they're supposed to.' Our model can tell us what their drug concentration is over time, and based on that, we can say whether the virus is growing or shrinking, and whether they're likely to develop resistance."

The model's predictions, Rosenbloom explained, can then serve as a guide to researchers as they work to design new drug cocktails to combat HIV.

While their model does hold out hope for simplifying the process of designing drug "cocktails," Hill and Rosenbloom said they plan to continue to refine the model to take additional factors -- such as multiple mutant-resistant strains of the virus and varying drug concentrations in other parts of the body -- into effect.

"The prototype we have so far looks at concentrations of drugs in blood plasma," Rosenbloom explained. "But a number of drugs don't penetrate other parts of the body, like the brains or the gut, with the same efficiency, so it's important to model these other areas where the concentrations of drugs might not be as high."

Ultimately, though, both say their model can offer new hope to patients by helping doctors design better, cheaper and more efficient treatments.

"Over the past 10 years, the number of HIV-infected people receiving drug treatment has increased immensely," Hill said. "Figuring out what the best ways are to treat people in terms of cost effectiveness, adherence and the chance of developing resistance is going to become even more important."
